IRIS SPE Method Development *Polymeric gels provide larger surface area that requires larger solvent volumes per gram of sorbent. Elution volumes depend on the chemical nature of the analyte, its relative concentration in the matrix, the chemistry of the solution solvent and the sorbent bed mass. This table outlines a generic SPE method. Choosing the Right Cartridges Cartridge Size —Sample Capacity Sizes: 1 mL, 3 mL, 6 mL, 60 mL (Specialty sizes available upon request) The SPE Cartidge determines the volume of the sample solution, which can be loaded onto the sorbent bed in a single step. Pore Size—the mean diameter of the pores on the IRIS 100% Divinyl Benzene Bead 104Å sorbent particle surface IR5424M6CX 60mg/3.0mL The analyte molecule in an SPE method should fit easily through the pores of the sorbent (i.e. for large molecules, a sorbent particle with extra large pores should be selected). Resin Bed Mass—Analyte Capacity Sizes: 10 mg, 30 mg, 60 mg, 200 mg, 1 g (Specialty bed weights available upon request) The resin bed mass determines the maximum amount of analyte retained by the sorbent. Tips: Choosing the proper bed mass reduces leaching of sample components Tips for Choosing Proper Resin Mass Resin Bed Volume—Interstitial and Pore Volume Polymeric Sorbents—polymeric resins provide larger The resin bed volume controls the minimum solvent surface area than silica-based resins, offering higher volume in an SPE method. To successfully condition, capacity on the order of 10-15% of the resin mass. This wash and elute from the cartridge typically requires allows for the use of smaller bed volumes and improved solvent volumes 4 to 8 times that of the sorbent bed. detection limts. Using less than 4 to 8 sorbent bed volumes contributes to incomplete solvation and low, inconsistent recoveries. Ion Exchange Sorbents—the number of charged sites on the resin available to interact with the analyte Tips: Polymeric SPE products usually require 200-250 uL per 100 determines capacity. mg of sorbent. Tip: Choosing a slightly larger resin mass is advisable, since most SPE samples are loaded with contaminants and interferences Toll Free: 1-888-226-2775 E-mail: [email protected] Web: www.lifescience.ca 425

P3 Protein Precipitation Plates P3 Protein Precipitation Plates P3 is now commonly used as the protein precipitation plate of choice in many major pharmaceutical companies. • Dual frit design Optional drain cap and top cap mats are available for those • Pre-filter frit at 100µm traps large flocculant particles wishing to use vortex sample mixing. • Secondary frit traps fine protein particles at <10µm • Frits are hydrophobic/oleophobic. This plate is used when sample and acetronitrile are • Specially selected polypropylene for low extractables pre-mixed before being pipetted into the plate. Biological samples commonly contain proteins that interfere with downstream applications. The P3 plate uses the CRASH method, in which the protein is denatured with acetronitrile and the flocculant filtered out, allowing 96 samples to be handled at one time. The Protein Precipitation Plate, P3, is based on the industry standard MicroLute™ format, but with- out the chromatographic sorbent. Protein ʻcrashesʼ out of solution and precipitates directly in each well when acetonitrile is added, thus solving all common problems associated with the CRASH technique of protein clean-up. The novel dual frit, hydrophobically treated matrix means that there is no ʻwetting outʼ and leakage of the sample through the plate before the application of vacuum. MicroLute™ BioVyon Co-sinter products BioVyon™ Co-sinter products are made using a solid-state media created from an ultra-pure and highly modified polymeric material with the lowest levels of extractables and leachables. Our proprietary BioVyon™ technology allows high-purity silica resins to be supported in a matrix which provides a high surface area whilst reducing channelling through the column. As the porous material is co-sintered under high pressure and temperature with the BioVyon™ polymer matrix, the resultant frit or column is impervious to further compression and easily resists channelling of liquids, even at high flow rates. BioVyon C8 and C18 products Porvair Sciences have introduced MicroLute™ BioVyon™ Co-sintered C8 and C18 silica SPE 96-well plates for low volume assays. Offered in 96-well MicroLute™ plates as a 10mg per well loading of C8 or C18 suitable for low sample volume bioassay preps and clean ups. Syringe and Membrane Filters Filter Device Selection Consider the number of samples, the sample volume, and the filtering mechanism you prefer to use. Syringe Filters Filtration is achieved by pushing the sample through the membrane with a syringe or other luer-connection device. Syringe filters allow you to control the rate of flow, which can be critical with delicate samples. Syringe filters also allow you to filter into nearly any tube, vial, or column that represents the next step in your analysis. Centrifuge Filter Tubes Filtration is achieved by pulling the sample through the membrane with centrifugal force. This allows you to simultaneously filter as many samples as your centrifuge can hold, and captures your filtrate in a clean microtube. Pre-Cut Membrane Multiple diameters offer a wide range of sample capacities, up to multiple liters. You need a filtration apparatus, such as a filter funnel or other vacuum-driven filtration device. TSS Filters for Standard Methods 2540 C and 2540 D Ensure worry free audits by taking the guesswork out of method compliance Xenosep TSS Filters are specifically designed to ensure compliance with the filter preparation requirements of Standard Methods 2540 C (Total Dissolved Solids Dried at 180°C) and Standard Methods 2540 D (Total Suspended Solids at 103-105°C). We removed the guesswork typically associated with audit checklist question #6 by making the wrinkle side of the filter visibly different than the screen side. Now everyone can clearly see and easily determine which side needs to face up to ensure compliance. Wrinkle side faces up In addition to more distinguishable filter surface characteristics Screen side faces down shown at left, these 1.5 µm binder-free, fast flowing, fine porosity borosilicate glass microfiber filters have been opti- mized to eliminate pin holing and minimize fiber shedding; two quality issues known to affect other brands of TSS filters.
