Investigating the functional and morphological neurological alterations within violent schizophrenic and psychopathic patients Aliza Hammond Schizophrenia is a chronic neuropsychiatric illness characterized by a loss of connection with reality, which in turn impacts the way patients think, emote, and behave. Approximately 12% of schizophrenic patients possess intrinsic violent and criminal propensities; however, there is a paucity of scientific understanding regarding the neurological mechanisms that underlie these expressions of schizophrenia. In response to this dearth of knowledge, Aliza will be investigating neurological, functional, and morphological alterations associated with this population. Furthermore, Aliza hopes to research how these neurological alterations implicate psychiatric dysfunction, such as deficits in cognitive control, decision making, motor control, and emotion processing. She is also focusing on increased impulsivity and urgency, as well as how these dysfunctions preempt violence. Aliza predicts that with an increased repertoire of violent-patient-specific neurological biomarkers, clinicians could identify patients likely to have violent tendencies and prevent those patients from inflicting harm upon others or themselves. Along with physical deterioration, patients with ALS and their families may also experience psychological decline, often manifesting in the form of apathy and social changes like social marginalization. Given the gravity of an ALS diagnosis and its severity, researchers recommend patients and their families meet with a palliative care specialist who is trained to address symptoms, psychological needs, and planning for the future. However, many specialists report a lack of experience in caring for patients with complex neurological diseases like ALS. The intestinal microbiota in anorexia nervosa Ella Javorsky Anorexia nervosa (AN) is a chronic psychiatric disorder characterized by the restriction of food and energy intake followed by weight loss, weight phobia, and body image disturbance. Among the somatic complications that result from malnutrition and disordered eating, researchers have reported the presence of gut dysbiosis. Gut dysbiosis is an altered composition of the intestinal microbiota associated with a decrease in the diversity of microorganisms. A relationship between the microbiota and psychopathology traits has been proposed, linking microbial dysbiosis to depression and anxiety, both of which are associated with anorexia nervosa. Thus, Ella would like to explore this connection in further depth and analyze the microbiota of AN patients throughout renourishment and treatment to evaluate how gut dysbiosis progresses with recovery. Ultimately, Ella hopes her future research will shed light on new treatments for AN by targeting microbial modulation during therapies. Excess sound energy can be converted into electrical energy, a more useful energy form. This energy conversion occurs via a piezoelectric transducer, which intrinsically transforms mechanical stress into electricity. In an audio harvesting system, a Helmholtz resonator is used to increase the sound level at a specific resonant frequency. A Helmholtz resonator is a cavity of air connected to a small neck, which works as a mass-spring system to cause this phenomenon. This increases the energy output, allowing the harvester to more efficiently power electronic devices. Ethan is looking to repurpose audio waste using an energy harvester to power small electronic devices. Exploring the neglected field of using sound as an energy source may help to address both noise pollution issues and offer a novel opportunity for renewable energy harvesting. Exploring how social media platforms can be used to reduce mental illness stigma and increase mental health literacy Madison Lee Mental illness stigma (MIS) causes discrimination due to an individual either having or being associated with a mental illness. MIS affected 20% of adults in the U.S. as of 2019, and the number of young adults with mental disorders is even higher. According to the National Institute of Mental Health, 49.5% of young adults 13-18 years old have a mental disorder. Most do not receive treatment due to lack of knowledge about their disorder, help-seeking stigma, or lack of resources available. In addition, many young adults today spend a large amount of time on social media platforms, and those platforms have been found to highly impact user beliefs. Madison is planning to look at these platforms to observe how social media influences MIS. Using these observations, she hopes to explore ways to decrease MIS and increase mental health literacy. The field of visual imagery research is relatively new, which means there is a great deal yet to be explored. The recent finding of individuals who lack the ability to see mental images altogether, aphantasics, represents a huge step forward in visual imagery research. It is estimated that two to three percent of the population exhibits aphantasia, yet most of those people do not realize they have it. Curiously, this realization often occurs only when another person mentions visual images. Aphantasics help us learn more about our own visual images and the mental processes associated with them. Tyler is interested in exploring how aphantasia and visual memory impact cognitive processes. Replacing degenerated retinal pigment epithelial cells in the macula to treat age-related macular degeneration Remi Matza Age-related macular degeneration (AMD) is the leading cause of severe vision loss in the developed world and affects 10-20% of people ages 65 and older. AMD causes the deterioration of the retinal pigment epithelium (RPE) and eventually photoreceptor cells in the macula, leading to a loss of central vision. Lacking effective therapies to treat AMD, scientists have recently found that the use of stem cells to replace degenerated RPE cells in the macula is extremely promising in renourishing and protecting photoreceptors. This allows for not only restoration of lost vision, but also prevention of further vision loss in patients with AMD. Remi hopes to research how this therapy can be optimized by looking at factors including stem cell types, growth substrates, and transplantation methods. She aims to explore this further to determine which treatment is most effective and leads to the greatest improvement in central vision. Kashani John Hopkins University Wilmer Eye Institute Baltimore, MD Exploring the immune/inflammatory impact of traumatic brain injuries on the permeability of the blood-brain barrier through an in vitro model of the neurovascular unit Samantha Milewicz Traumatic brain injuries (TBI) are physical injuries to the brain from an external force, causing impairment. Each year, TBIs affect 1.7 million people, and due to their severity, damage may be irreversible. The blood-brain barrier (BBB), an essential part of the neurovascular unit (NVU), separates the brain’s microenvironment from circulation; however, a TBI can cause BBB disruption, putting the brain at increased risk for a cascade of negative events such as increased fluid buildup, edema, and neuronal cell damage. Further research is necessary to understand the contribution of such disturbances to BBB integrity. An ideal in vitro TBI model would mimic disease pathology through imposed mechanical, metabolic, and inflammatory insults on an NVU model. Under her mentor’s guidance, Samantha hopes to conduct a set of experiments related to altered physiological parameters on the NVU platform developed by her mentor, with an eye toward combined inflammatory/immune impact on BBB permeability. Exploring the effect of light therapy on the circadian molecular clock in patients with seasonal affective disorder Lindsay Miller Seasonal affective disorder (SAD) is a depressive disorder with a recurring seasonal pattern. Antidepressants have treated similar disorders for decades, but contemporary research suggests that light therapy may be the optimal solution for SAD patients. A lack of sunlight contributes to the development of SAD, and light therapy can extend a patient’s photoperiod, or daily exposure to light. This artificially lengthened photoperiod corrects the dysfunction of the sleep/wake cycle, which is controlled by the expression of a set of genes. Light therapy has an altering effect on the expression of these genes, so Lindsay aims to further explore this relationship. Understanding the genetic basis for this disease may open up the possibility of new targets for treatment. Lindsay hopes to further investigate light therapy’s effect on the genes that contribute to the severity of SAD, which could ultimately minimize the detrimental impacts of the disorder as a whole. Because of this, Peter is interested in the application of game- theoretic models for fighting cancer and how these models can be used to predict behaviors of cancer cells. These predictions could ultimately lead to the use of specific cancer treatments based on how cancer cells are anticipated to behave. Peter hopes to create his own game-theoretic model to envision the interactions between individual cancer cells to find an exploitable behavior. If found, such a behavior could be used in favor of the physician and patient in ways that include finding an optimal time for treatment or an ideal treatment type. Ultimately, Peter wants to find a specific behavior that would be greatly advantageous to physicians in combating cancer. CAR T cell therapy in acute myeloid leukemia Kaela Newman Acute myeloid leukemia (AML) is the most common form of leukemia in adults, and although remission can be achieved through treatment, the five- year survival rate post-therapy is 28.3%. The current standard treatment for AML does not eradicate leukemia stem cells (LSCs), the cells that cause the disease. It is imperative for AML treatment to utilize a mechanism of specific cell targeting so that LSCs can be eradicated. Chimeric antigen receptor (CAR) T cells allow for the specific targeting of cells and show promise in lymphoid malignancies. Kaela hopes to study the implementation of CAR T cells in AML. She specifically aims to research the targeting of antigen CD123, which is expressed on LSCs and has lower toxicity to normal hematopoietic stem and progenitor cells (HSPCs) compared with other targets. Ultimately, she hopes her work will shed light on the potential efficacy of CAR T cell therapy in AML. Specifically, brain cancer has one of the lowest survivability rates of any cancer. The conventional treatment for brain cancers is resection, wherein neurosurgeons remove a critical mass of the tumor. Maximal tumor resection is essential to patient survivability, as it prevents cancer recurrence. However, even with advanced intraoperative imaging techniques, such as stimulated Raman histology (SRH), maximal tumor resection is difficult to achieve. Thus, there is a demand for rapid, automated brain tumor diagnosis that surgeons can use in situ. Convolutional neural networks (CNNs) are neural network architectures used for image recognition. Caleb is interested in designing CNNs that generate accurate guides by analyzing tumor images made using an SRH microscope. This could improve the efficacy of surgical resection treatment by increasing the likelihood of maximal tumor resection. Utilizing focused ultrasound with microbubbles to non-invasively open the blood-brain barrier and permit cancer treating drugs to reach the brain Samantha Schaevitz Approximately 80,000 people in the United States will be diagnosed this year with a brain tumor. Although treatment of brain cancers has vastly improved over the years, scientists are still far from finding a cure. Samantha is interested in the use of a novel technique, focused ultrasound with microbubbles, to open the blood-brain barrier and allow cancer treatment drugs to reach the brain. Ultrasound frequencies cause microbubbles within the barrier to oscillate, which opens tight junctions that form the barrier around the brain. Once open, drugs that were previously blocked can then reach the brain, allowing for more effective treatment. Studies have shown this process to result in successful opening of the barrier with no neurological deficits post-treatment. Samantha is excited to continue her research with her mentor at Columbia University and to delve deeper into drug delivery within brain tumors. Wes plans to focus his study on the synthesis of monolayer, bilayer, and multilayer graphene for these specific applications. Graphene forms through the process of chemical vapor deposition (CVD). During the CVD process, methane is allowed into a chamber at ~1,323 K, the temperature at which methane breaks apart into hydrogen and carbon. The carbon then rearranges itself and settles onto a copper surface in a 2D hexagonal formation called graphene. Sometimes the graphene sheets form with imperfections that can hinder the desired electrical or mechanical properties of the graphene. Utilizing the bowhead whale’s (Balaena mysticetus) genome and transcriptome to search for pro-longevity genes Emily Stangel Many species have drastically different lifespans, but a definitive molecular explanation for this remains unknown. Interestingly, the bowhead whale (Balaena mysticetus) can live up to 200 years. In order to determine the molecular mechanisms responsible for the increased longevity of bowhead whales, researchers analyzed their genome and transcriptome to locate sequences associated with DNA repair, cell-cycle regulation, and aging. To accurately find these sequences, the bowhead whale’s DNA and RNA were compared to that of other well-researched mammals such as mice, naked mole-rats, and other Cetaceans. These organisms rarely contract diseases that usually coincide with aging, such as cancer. Emily is interested in how the bowhead whale’s pro- longevity genes decrease the risk of disease with increased xenobiotic factors in their environments. By locating genes associated with increased longevity in mammalian species, drugs could be developed to mimic, in humans, the benefits conferred by these genes. Vascular malformations and tumors appear most often at birth or in early childhood. Due to their extreme rarity, vascular anomalies are considerably understudied. For the rarest of anomalies, such as kaposiform hemangioendothelioma (KHE), diagnosis is arduous, and treatment is dominated by therapies that lack long-term efficacy, often coming with serious side effects. As a result, researchers are looking at the underlying genetic mutations and mechanisms associated with these anomalies to improve diagnosis, management, and overall treatment. Max hopes to research these mutations and novel treatments. Specifically, he aims to further explore these rare malformations and tumors. He plans to work under the American Society of Pediatric Hematology-Oncology to improve the understanding of risk factors, underlying molecular changes, and treatment response in children with KHE. - Contextual Information Regarding Awards - The Regeneron Science Talent Search The Regeneron Science Talent Search is “the Nation’s Oldest and Most Prestigious Science and Mathematics Competition. Started in 1942 as the Westinghouse Science Talent Search, the Regeneron Science Talent Search (Regeneron STS) recognizes and empowers our nation’s most promising young scientists who are developing ideas that could solve society’s most urgent challenges. Each year, nearly 1,900 students enter the Regeneron STS, submitting original research in critically important scientific fields of study. Unique among high school competitions, the Regeneron Science Talent Search focuses on identifying, inspiring, and engaging the most promising scientists among the nation’s high school seniors” (societyforscience.org). The Junior Science and Humanities Symposia The Junior Science and Humanities Symposia (JSHS) Program is “a tri-service– U.S. Army, Navy, and Air Force-sponsored STEM competition which promotes original research and experimentation in the sciences, technology, engineering, and mathematics (STEM) at the high school level and publicly recognizes students for outstanding achievement. By connecting talented students, their teachers, and research professionals at affiliated symposia and by rewarding research excellence, JSHS aims to widen the pool of trained talent prepared to conduct research and development vital to our nation. Endorsed by the National Association of Secondary School Principals (NASSP), JSHS regional and national symposium are held during the academic year and reach more than 8,000 high school students and teachers throughout the United States, Puerto Rico, and the Department of Defense Schools of Europe and the Pacific Rim. Students must first participate in their regional symposia where they compete for selection to present at the national symposium each year” (jshs.org). The Westchester Science and Engineering Fair WESEF, the Westchester Science and Engineering Fair, “offers high school students from Westchester and Putnam counties an opportunity to present their research to scientists, business leaders, and the general public.
