EDITORIAL Hope is not simply being optimistic or happy all the time. To have hope is to want an outcome that makes your life better in some way. It not only can help make a tough present situation more bearable but also can eventually improve our lives because envisioning a better future motivates you to take the steps to make it happen.Hope is not the lottery of luck or the hope some students have for higher grades when they have not completed their assignments. That may be wishful thinking but not hopeful thinking. Whether we think about it or not, hope is a part of everyone’s life. Everyone hopes for something. It’s an inherent part of being a human being. Hope helps us define what we want in our futures and is part of the self- narrative about our lives we all have running inside our minds. -J.R.R. Tolkien: “Oft hope is born when all is forlorn.”

12 AGAR JELLY Agar or agar-agar, is a jelly-like substance consisting of polysaccharides obtained from the cell walls of some species of red algae, primarily from ogonori (Gracilaria) and \"tengusa\" (Gelidiaceae). Agar is a mixture of two components, the linear polysaccharide agarose and a heterogeneous mixture of smaller molecules called agaropectin. It forms the supporting structure in the cell walls of certain species of algae and is released on boiling. These algae are known as agarophytes, belonging to the Rhodophyta (red algae) phylum. Agar has been used as an ingredient in desserts throughout Asia and also as a solid substrate to contain culture media for microbiological work. Agar can be used as a laxative; an appetite suppressant; a vegetarian substitute for gelatin; a thickener for soups; in fruit preserves, ice cream, and other desserts; as a clarifying agent in brewing; and for sizing paper and fabrics. Dhokla While making dhokla add besan {gram flour from chickpeas} ,Then add to it lemon juice and curd (this helps in adding required acid and lactobacilli) Fermentation of batter is carried out by Lactobacillus or lactic acid bacteria. These bacteria with oxygen in air and sugar/starch they form carbon-dioxide, water and lactic acid. Water dissolves in batter and carbon dioxide forms bubbles and batter swells. This carbon dioxide should be trapped in batter Fermented foods are easier to digest. This helps enable a healthy gut flora and an improved digestion.

14 DEPARTMENT OF MICROBIOLOGY (ID) Mushroom Cultivation mushroom, the conspicuous umbrella-shaped fruiting body (sporophore) of certain fungi, typically of the order Agaricales in the phylum Basidiomycota but also of some other groups. Popularly, the term mushroom is used to identify the edible sporophores; the term toadstool is often reserved for inedible or poisonous sporophores. There is, however, no scientific distinction between the two names, and either can be properly applied to any fleshy fungus fruiting structure. In a very restricted sense, mushroom indicates the common edible fungus of fields and meadows (Agaricus campestris). A very closely related species, A. bisporus, is the mushroom grown commercially and seen in markets. Umbrella-shaped sporophores are found chiefly in the agaric family (Agaricaceae), members of which bear thin, bladelike gills on the undersurface of the cap from which the spores are shed. The sporophore of an agaric consists of a cap (pileus) and a stalk (stipe). The sporophore emerges from an extensive underground network of threadlike strands (mycelium). An example of an agaric is the honey mushroom (Armillaria mellea). Mushroom mycelia may live hundreds of years or die in a few months, depending on the available food supply. As long as nourishment is available and temperature and moisture are suitable, a mycelium will produce a new crop of sporophores each year during its fruiting season. Fruiting bodies of some mushrooms occur in arcs or rings called fairy rings. The mycelium starts from a spore falling in a favourable spot and producing strands (hyphae) that grow out in all directions, eventually forming a circular mat of underground hyphal threads. Fruiting bodies, produced near the edge of this mat, may widen the ring for hundreds of years.
15 DEPARTMENT OF MICROBIOLOGY (ID) A few mushrooms belong to the order Boletales, which bear pores in an easily detachable layer on the underside of the cap. The agarics and boletes include most of the forms known as mushrooms. Other groups of fungi, however, are considered to be mushrooms, at least by laymen. Among these are the hydnums or hedgehog mushrooms, which have teeth, spines, or warts on the undersurface of the cap (e.g., Dentinum repandum, Hydnum imbricatum) or at the ends of branches (e.g., H. coralloides, Hericium caput-ursi). The polypores, shelf fungi, or bracket fungi (order Polyporales) have tubes under the cap as in the boletes, but they are not in an easily separable layer. Polypores usually grow on living or dead trees, sometimes as destructive pests. Many of them renew growth each year and thus produce annual growth layers by which their age can be estimated. Examples include the dryad’s saddle (Polyporus squamosus), the beefsteak fungus (Fistulina hepatica), the sulfur fungus (P. sulphureus), the artist’s fungus (Ganoderma applanatum, or Fomes applanatus), and species of the genus Trametes. The clavarias, or club fungi (e.g., Clavaria, Ramaria), are shrublike, clublike, or coral-like in growth habit. One club fungus, the cauliflower fungus (Sparassis crispa), has flattened clustered branches that lie close together, giving the appearance of the vegetable cauliflower. The cantharelloid fungi (Cantharellus and its relatives) are club-, cone-, or trumpet-shaped mushroomlike forms with an expanded top bearing coarsely folded ridges along the underside and descending along the stalk. Examples include the highly prized edible chanterelle (C. cibarius) and the horn-of-plenty mushroom (Craterellus cornucopioides). Puffballs (family Lycoperdaceae), stinkhorns, earthstars (a kind of puffball), and bird’s nest fungi are usually treated with the mushrooms. The morels (Morchella, Verpa) and false morels or lorchels (Gyromitra, Helvella) of the phylum Ascomycota are popularly included with the true mushrooms because of their shape and fleshy structure; they resemble a deeply folded or pitted conelike sponge at the top of a hollow stem. Some are among the most highly prized edible fungi (e.g., Morchella esculenta). Another group of ascomycetes includes the cup fungi, with a cuplike or dishlike fruiting structure, sometimes highly coloured.
Microbiology 16 DEPARTMENT OF MICROBIOLOGY (ID) Lab Practices and Safety Rules 1. Wash your hands with disinfectant soap when you arrive at the lab and again before you leave. 2. Absolutely no food, drinks, chewing gum, or smoking is allowed in the laboratory. Do not put anything in your mouth such as pencils, pens, labels, or fingers. Do not store food in areas where microorganisms are stored. 3. Purchase a lab coat and safety glasses, bring them to class, and use them. Alternatively, a long sleeved shirt that buttons or snaps closed is acceptable protective clothing. This garment must cover your arms and be able to be removed without pulling it over your head. Leave protective clothing in the lab and do not wear it to other non-lab areas. 4. Avoid loose fitting items of clothing. Wear appropriate shoes (sandals are not allowed) in the laboratory. 5. Keep your workspace free of all unnecessary materials. Backpacks, purses, and coats should be placed in the cubbyholes by the front door of the lab. Place needed items on the floor near your feet, but not in the aisle.
17 6. Disinfect work areas before and after use with 70% ethanol or fresh 10% bleach. Laboratory equipment and work surfaces should be decontaminated with an appropriate disinfectant on a routine basis, and especially after spills, splashes, or other contamination. 7. Label everything clearly. 8. Replace caps on reagents, solution bottles, and bacterial cultures. Do not open Petri dishes in the lab unless absolutely necessary. 9. Inoculating loops and needles should be flame sterilized in a Bunsen burner before you lay them down. 10. Turn off Bunsen burners when not is use. Long hair must be restrained if Bunsen burners are in use. 11. When you flame sterilize with alcohol, be sure that you do not have any papers under you. 12. Treat all microorganisms as potential pathogens. Use appropriate care and do not take cultures out of the laboratory. 13. Wear disposable gloves when working with potentially infectious microbes or samples (e.g., sewage). If you are working with a sample that may contain a pathogen, then be extremely careful to use good bacteriological technique. 14. Sterilize equipment and materials. 15. Never pipette by mouth. Use a pipetting aid or adjustable volume pipettors. [In the distant past, some lab personnel were taught to mouth pipette. This practice has been known to result in many laboratory- acquired infections. With the availability of mechanical
18 16. Consider everything a biohazard. Do not pour anything down the sink. Autoclave liquids and broth cultures to sterilize them before discarding. 17. Dispose of all solid waste material in a biohazard bag and autoclave it before discarding in the regular trash. 18. Familiarize yourself with the location of safety equipment in the lab (e.g., eye-wash station, shower, sinks, fire extinguisher, biological safety cabinet, first aid kit, emergency gas valve). 19. Dispose of broken glass in the broken glass container. 20. Dispose of razor blades, syringe needles, and sharp metal objects in the “sharps” container. 21. Report spills and accidents immediately to your instructor. Clean small spills with care (see instructions below). Seek help for large spills. 22. Report all injuries or accidents immediately to the instructor, no matter how small they seem.
Know your 19 MICROBES DEPARTMENT OF Food biotechnology is the application of modern biotechnological MICROBIOLOGY (ID) techniques to the manufacture and processing of food products as well Quiz Highlights as food ingredients and food additives. Although agricultural biotechnology, such as crop breeding and animal production, are generally regarded as food biotechnology, it is expected to have a great impact on the agri-food industry. The food industry has gone through major changes in recent years. These changes are in response to consumers’ demand for food products low in calories and made with natural ingredients and additives, as opposed to synthetic ones. The overall efforts are directed for the development of novel approaches to produce food products that will meet the consumer's demand for healthy foods (Sanders, 1991). There is a spectacular increase in the use of enzymes as useful tools in the food industry (Singh et al., 2019). These applications cover a wide range of biotechnological applications as well as food production, food processing, and food preservation (Raveendran et al., 2018). There are currently amazing new developments in fermentation, enzyme technology, genetic engineering, bioengineering, and food processing. Although biotechnology may involve the use of one or many living forms, microorganisms have an important and significant role in its development, due to their rapid production of large amounts of biomasses (Vasic-Racki et al., 2011). In addition, such production could be obtained with an added-value approach by using inexpensive culture media of agri-food waste and residual materials (Bull, 1983). Using microorganisms, the innovations in genetic engineering (Perkel, 2019) contributed greatly to the development of many desirable enzymes for the food industry (Smith and Shaw, 2008). Different bioconversion processes are carried out by enzymes, including de novo synthesis throughout precursor fermentation as well as single and multistep bioconversions. However, most of enzymes of interest in food industry are often extracellular rather than intracellular. Overall, research in food biotechnology is mainly focused on the improvement of enzymes, currently used by the industry, in terms of their stabilities and specificities at different stages of the processes (Raveendran et al., 2018). In addition, the development of novel and unique enzymes as well as their industrial applications is a race for knowledge. With environment and economic concerns regarding agri- food wastes and residues, there is increasing interest in the development of novel approaches using a single enzyme or multienzyme system in bioprocessing for the bioconversion of desirable added-value products
20 DEPARTMENT OF MICROBIOLOGY (ID) Advantages of Quizzes Students tend to actually read the material. Students show up for class on time since the quiz always comes first. Students are placed in the right attitude for learning. Students feel more confident to discuss the material. Students raise their grades by simply reading the material. The start of class is signaled. The quiz provides a good lead-in for either a lecture or discussion of the material. Students grow curious about the answers. At the end of class (when the suspense is killing them), I usually answer any quiz question that didn't naturally pop up during class. The easy chance to do well gives students a feeling of self-esteem. 40 quizzes can be graded in about five minutes, and I don't feel a need to ask factual questions on tests just to see if they've read the material. Students are provided with a real foundation for intellectual growth (How can a student truly learn to interpret a work without knowing the facts? Note the case study method in law school depends on the students first grasping the essential facts of the case).
